Pucotenimab (HX008) is a humanized IgG4 monoclonal antibody targeting the programmed cell death protein 1 (PD-1) receptor. Developed by Lepu Biopharma (Hanx Biopharmaceuticals), it acts as an immune checkpoint inhibitor by blocking the interaction between PD-1 and its ligands, PD-L1 and PD-L2. This blockade prevents the down-regulation of T-cell activity, thereby restoring and enhancing the body's anti-tumor immune response. Pucotenimab has received approval from the National Medical Products Administration (NMPA) in China for the treatment of patients with unresectable or metastatic melanoma who have failed prior systemic therapy, as well as for advanced solid tumors characterized by high microsatellite instability (MSI-H) or mismatch repair deficiency (dMMR). It is also being investigated in combination therapies, such as with cryoablation or radiotherapy, for managing cancer-related pain in patients with bone metastases.
