Pucotenlimab is a recombinant humanized anti-PD-1 monoclonal antibody of the IgG4/kappa subtype. It selectively blocks the binding of programmed cell death protein 1 (PD-1) with its ligands PD-L1 and PD-L2, thereby restoring immune cell activity against cancer cells. Developed by Lepu Biopharma Co., Ltd., it received its first global approval in China in July 2022 for the treatment of unresectable or metastatic microsatellite instability-high (MSI-H) or mismatch repair deficient (dMMR) solid tumors. It has also been approved for unresectable or metastatic melanoma after failure of previous systemic therapy[4][7][10]. Pucotenlimab is being investigated for use in other cancers.
