Pucotenlimab + becotatug vedotin is a combination regimen consisting of becotatug vedotin (MRG003), an epidermal growth factor receptor (EGFR)-targeted antibody-drug conjugate (ADC), and pucotenlimab (HX008), a monoclonal antibody targeting the programmed cell death protein 1 (PD-1). Becotatug vedotin comprises a humanized anti-EGFR antibody conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE) via a protease-cleavable valine-citrulline linker. The combination is designed to leverage the direct cytotoxic effects of the ADC against EGFR-expressing tumor cells and the immune-restoring properties of the PD-1 inhibitor. It is currently being evaluated in Phase III clinical trials as a maintenance therapy for patients with recurrent or metastatic nasopharyngeal carcinoma (NPC) who have achieved stable disease or have positive plasma EBV DNA following first-line chemoimmunotherapy.
