Overview

PUL-042 is a first-in-class, inhaled immunomodulatory agent composed of synthetic ligands for Toll-like receptor (TLR) 2/6 and TLR 9. It is designed to stimulate the lung’s innate immune system by activating TLRs on lung epithelial cells, leading to the release of antimicrobial peptides and reactive oxygen species that inhibit and kill a wide range of respiratory pathogens. This activation also triggers an adaptive immune response. Developed primarily as a broad-spectrum therapy for respiratory infections, including COVID-19, MERS, SARS, and other viral or bacterial pathogens, PUL-042 has demonstrated efficacy in preclinical models—even in immunocompromised animals—and has shown favorable tolerability in clinical trials. The drug is being developed by Pulmotect.
