Drug intelligence / Profile preview
Pulmazole (PUR1900) is an inhaled dry powder formulation of the triazole antifungal itraconazole, engineered using Pulmatrix's proprietary iSPERSE (inhaled Small Particles Easily Respirable and Emitted) technology. iSPERSE utilizes small, dense, and dispersible particles to ensure efficient delivery to the airways via a dry powder inhaler. The drug is primarily being developed for the treatment of Allergic Bronchopulmonary Aspergillosis (ABPA) in patients with asthma or cystic fibrosis. By delivering itraconazole directly to the site of fungal colonization in the lungs, Pulmazole aims to achieve therapeutic local concentrations while significantly reducing systemic exposure. This approach is intended to mitigate the risks of systemic toxicities and complex drug-drug interactions (mediated by CYP3A4 inhibition) typically associated with oral itraconazole therapy. The mechanism of action involves the inhibition of fungal lanosterol 14-alpha-demethylase (CYP51), which prevents the conversion of lanosterol to ergosterol, thereby compromising fungal cell membrane integrity and inhibiting growth.
