Pumarix is an adjuvanted, inactivated, split-virion pandemic influenza vaccine developed by GSK Biologicals. It is specifically designed to provide immunization against the H5N1 subtype of influenza A virus. The vaccine comprises purified antigen fractions of the inactivated H5N1 virus, formulated with GSK's proprietary AS03 adjuvant system, which is an oil-in-water emulsion containing squalene, DL-α-tocopherol, and polysorbate 80. By utilizing this adjuvant, the vaccine is able to induce a more robust and broader immune response with a lower dose of antigen, a concept known as dose-sparing, which is critical during pandemic scenarios. The vaccine was granted marketing authorization in Europe in 2011 for use in officially declared pandemic situations, though the authorization was later withdrawn in 2015 for commercial reasons unrelated to safety or efficacy.
