pUMVC3 is a mammalian expression plasmid vector backbone widely utilized in gene therapy and DNA vaccine research. Originally developed as a retroviral packaging plasmid (often associated with Bob Weinberg's lab at the Whitehead Institute and the University of Michigan Vector Core), pUMVC3 contains a cytomegalovirus (CMV) promoter/enhancer and is designed for high-level expression of transgenes in mammalian cells. In addition to its role as a cloning vector for therapeutic genes (such as IL-12 or tumor-associated antigens), the empty pUMVC3 plasmid has been shown to possess intrinsic immunostimulatory properties. When delivered into the cytoplasm of tumor cells via electroporation, the exogenous plasmid DNA acts as a danger signal, upregulating the surface expression of MHC class I (MHC-I) and PD-L1, thereby promoting local anti-tumor immune responses and attenuating tumor growth in preclinical models.
