pUMVC3-IGFBP-2 is a plasmid DNA vaccine encoding the human insulin-like growth factor-binding protein 2 (IGFBP-2) gene. Developed by researchers at the University of Washington, this therapeutic vaccine is designed to elicit a Th1-type immune response against tumor cells that overexpress IGFBP-2, a protein associated with tumor progression, metastasis, and treatment resistance in various malignancies. The vaccine utilizes the pUMVC3 mammalian expression vector to facilitate the intracellular expression of the IGFBP-2 antigen. Once expressed, the antigen is processed and presented by major histocompatibility complex (MHC) molecules, stimulating the activation of IGFBP-2-specific cytotoxic T-lymphocytes (CTLs) and helper T-cells. This immunotherapeutic approach specifically targets the oncogenic pathways mediated by IGFBP-2 in cancers such as HER2-negative breast cancer and high-grade serous ovarian cancer.
