Overview

Punalimab (formerly TJ-S2) is a humanized monoclonal antibody designed to target and neutralize interleukin-1 beta (IL-1β). Developed by I-Mab Biopharma, it is currently being evaluated in clinical trials for the treatment of acute gouty arthritis, particularly in patients who have contraindications, intolerance, or an inadequate response to conventional treatments such as nonsteroidal anti-inflammatory drugs (NSAIDs) and colchicine. By blocking IL-1β, punalimab interrupts the inflammatory cycle responsible for the intense pain and swelling associated with gout flares. The drug is being developed by I-Mab's Hangzhou subsidiary and is part of a broader strategy to address unmet needs in inflammatory and autoimmune diseases.
