PuraStat is a synthetic, transparent, self-assembling hydrogel peptide developed by 3-D Matrix, Ltd. Its active ingredient is RADA16, a linear 16-amino acid oligopeptide with a repeating sequence of arginine (R), alanine (A), aspartic acid (D), and alanine (A). Upon contact with blood or physiological fluids, the acidic peptide solution is neutralized and self-assembles into β-sheet nanofibers that form a three-dimensional hydrogel scaffold. This scaffold physically coats the bleeding site to achieve hemostasis. Classified as a medical device, PuraStat is bioinert and does not interact with the coagulation cascade. It is indicated for intraoperative hemostasis across cardiovascular, gastrointestinal (GI), and otorhinolaryngological surgical procedures, as well as for endoscopic control of GI bleeding (including non-variceal bleeding and bleeding associated with procedures such as polypectomy and ESD) and for the prevention of delayed postoperative bleeding.
