Overview

PIZV (purified inactivated Zika virus vaccine), also known as TAK-426, is a candidate prophylactic vaccine developed to protect against infection by the Zika virus. It is a purified, formalin-inactivated whole-virus preparation of the PRVABC59 strain of ZIKV and is adjuvanted with aluminum hydroxide. The vaccine is administered intramuscularly in two doses 28 days apart. Clinical studies have shown that PIZV elicits robust and persistent neutralizing antibody responses comparable to those seen after natural infection, with an acceptable safety profile in both flavivirus-naive and flavivirus-primed adults[1][2][6]. The primary mechanism of action involves induction of protective immunity through generation of neutralizing antibodies against the Zika virus envelope proteins.
