Overview

**Puromycin aminonucleoside** is the aminonucleoside portion of the antibiotic puromycin, structurally defined as 3'-amino-3'-deoxy-N6,N6-dimethyladenosine. It is primarily used as a research tool in nephrology, most notably to induce nephrotic syndrome/nephrosis in animal (especially rat) models by damaging podocytes, the specialized glomerular epithelial cells required for kidney filtration. The compound interferes with protein synthesis and terminates ribosomal peptide chain elongation, resulting in inhibition of translation and triggering of apoptosis via endoplasmic reticulum (ER) stress pathways in podocytes. Mechanistically, puromycin aminonucleoside causes ER stress involving ATF6α, GRP78/BiP, and caspase-12, leading to podocyte apoptosis and features of glomerular disease such as proteinuria[1][2][3][5]. Its applications include studying mechanisms of glomerular disease, screening nephroprotective compounds, and research into protein synthesis inhibitors and antineoplastic mechanisms[2][5].
