Overview

**Purpurin 18** is a second-generation photosensitizer derived from chlorophyll, belonging to the chlorin class of compounds. It is utilized primarily in **photodynamic therapy (PDT)** for cancer treatment. Upon activation by light (wavelengths >650 nm), purpurin 18 produces reactive oxygen species, notably singlet oxygen, resulting in targeted oxidative damage to cancer cells while sparing normal tissue. Its mechanism of action centers on photodynamic generation of cytotoxic species and induction of apoptosis in tumor cells. Purpurin 18's advantages include a long absorption spectrum, high singlet oxygen quantum yield, and low dark toxicity. Major limitations are its hydrophobicity, poor water solubility, tendency to aggregate, and limited tumor selectivity—these are being addressed through formulations such as PEGylation and solid lipid nanoparticles to improve bioavailability and therapeutic effect.
