The Puumala virus DNA vaccine is an investigational DNA-based vaccine designed to prevent Hemorrhagic Fever with Renal Syndrome (HFRS) or Nephropathia epidemica caused by the Puumala virus (PUUV). It utilizes the pWRG7077 plasmid backbone to deliver the M segment of the Puumala virus genome, which encodes the viral envelope glycoproteins (Gn and Gc). Following intramuscular administration—often facilitated by electroporation or needle-free jet injection—the plasmid enters host cells and expresses these glycoproteins, which then serve as antigens to stimulate a protective immune response, including the production of neutralizing antibodies. This vaccine is frequently evaluated both as a monotherapy and as part of a multivalent formulation with the Hantaan virus DNA vaccine to provide broad protection against major hantaviruses endemic in Eurasia.
