Puveitinib (DR-106) is an orally bioavailable, small molecule multi-kinase inhibitor developed by Hangzhou Derun Yucheng Biotechnology. It is designed to simultaneously target three critical pathways in oncology: angiogenesis, tumor cell proliferation, and the immune microenvironment. Specifically, puveitinib inhibits Vascular Endothelial Growth Factor Receptors (VEGFR 1/2/3), Fibroblast Growth Factor Receptors (FGFR 1/2/3), and the Colony-Stimulating Factor 1 Receptor (CSF-1R). By blocking VEGFR and FGFR, the drug suppresses tumor-associated blood vessel formation and direct oncogenic signaling. Its inhibition of CSF-1R is intended to deplete or reprogram immunosuppressive tumor-associated macrophages (TAMs), thereby enhancing the anti-tumor immune response. Puveitinib is currently in Phase 1 clinical development for the treatment of advanced solid tumors.
