PV-10 is a small molecule cancer immunotherapy whose active ingredient is rose bengal disodium. It is designed for intralesional injection into solid tumors and acts through dual mechanisms of local tumor ablation and systemic immunological activation. Mechanistically, PV-10 induces direct cytotoxicity in tumor cells via apoptosis and necrosis, as well as immune-mediated anti-tumor effects. Preclinical studies have shown that PV-10 activates the STING pathway (stimulator of interferon genes), leading to the release of interferon gamma and enhanced immune response[3][6]. Clinically, it has been investigated primarily for advanced cutaneous melanoma (including metastatic melanoma) but also in other solid tumors such as hepatocellular carcinoma and uveal melanoma metastatic to the liver[1][2][5][7]. The drug has received orphan drug designation from the FDA for metastatic melanoma and hepatocellular carcinoma[5][7]. Developed by Provectus Biopharmaceuticals, it has demonstrated promising response rates both as monotherapy and in combination with immune checkpoint inhibitors like pembrolizumab (Keytruda)[2][4].
