Overview

**pVAX1 + C-G250** is a combination DNA vaccine candidate for renal cell carcinoma (kidney cancer). It consists of two components: the pVAX1 plasmid backbone encoding the tumor-associated antigen G250 (carbonic anhydrase IX, CAIX, also known as C-G250), and the pVAX1 plasmid vector as the delivery system. The vaccine works by stimulating both humoral (antibody-mediated) and cellular (T cell-mediated) immune responses against tumor cells expressing the G250 antigen, thereby enhancing anti-tumor immunity. In preclinical studies, the addition of a protein boost (such as C-G250 protein) following the DNA prime further potentiates the immune response. Polyethyleneimine (PEI) has been used as a delivery enhancer in these formulations, and intramuscular administration followed by electroporation is used to optimize DNA uptake and expression in vivo. The vaccine is designed to treat renal cell carcinoma by targeting tumor cells for immune-mediated destruction through antigen-specific antibody and T-cell responses[3][5][1].
