pVAX1-6Aβ15-T is an experimental DNA epitope chimeric vaccine designed for the treatment and prevention of Alzheimer's disease. Developed by researchers at the Beijing Institute of Biotechnology, the vaccine consists of a pVAX1 plasmid vector containing a synthetic chimeric minigene. This minigene encodes six tandem copies of the immunodominant B-cell epitope Aβ1-15 (the first 15 amino acids of amyloid-beta) linked by GS linkers and fused with the Pan HLA-DR reactive epitope (PADRE), a universal T-helper epitope. The vaccine aims to elicit high titers of anti-Aβ antibodies that recognize Aβ oligomers and plaques while avoiding the harmful T-cell-mediated inflammatory responses associated with full-length Aβ42 vaccines. Preclinical studies in mouse models (such as PDAPP mice) have demonstrated its ability to reduce cerebral amyloid load and improve cognitive function, particularly when used in a co-immunization strategy with its corresponding recombinant protein vaccine.
