Drug intelligence / Profile preview
PvCSP nanoparticle vaccine (PvCSP NPV) is a pre-erythrocytic stage malaria vaccine candidate designed to prevent infection by the parasite *Plasmodium vivax*. Developed collaboratively by the University of South Florida, Johns Hopkins University, and the University of Florida, the vaccine utilizes a biodegradable poly(lactic-co-glycolic acid) (PLGA) nanoparticle platform fabricated via flash nanoprecipitation (FNP) to produce uniform particles of less than 100 nm. The vaccine co-delivers recombinant *P. vivax* circumsporozoite protein (PvCSP) subdomains—such as full-length (CSPFL), N-terminal (CSPNT), or C-terminal (CSPCT) domains—surface-conjugated to the PLGA nanoparticles alongside the Toll-like receptor 9 (TLR9) agonist adjuvant CpG-1018. In preclinical mouse models, the PvCSP NPV elicits high-titer, neutralizing anti-CSP antibodies that recognize native sporozoite antigens, inhibit hepatocyte invasion, and provide protection against sporozoite challenge.
