PVP-TZ-POR is an experimental photodynamic therapy (PDT) formulation consisting of novel beta-substituted triazole-porphyrin derivatives (TZ-PORs) encapsulated within polyvinylpyrrolidone (PVP) micelles. The PVP carrier is utilized to enhance the water solubility and prevent the aggregation of the hydrophobic porphyrin photosensitizers, which is critical for maintaining their photophysical efficiency in physiological environments. Upon exposure to light (typically in the visible spectrum), the TZ-POR molecules undergo electronic excitation and transfer energy to molecular oxygen, generating highly reactive singlet oxygen (1O2). This localized production of reactive oxygen species (ROS) leads to oxidative stress and subsequent apoptosis or necrosis of target cells. Preclinical in vitro studies have demonstrated that PVP-TZ-POR formulations, particularly those containing derivatives 7e and 7f, exhibit high phototoxicity against human bladder cancer cell lines (HT-1376) while showing minimal effect on normal epithelial cells (ARPE-19).
