Drug intelligence / Profile preview
Pvs230D1-EPA is a transmission-blocking vaccine (TBV) candidate for *Plasmodium vivax* malaria. It consists of the recombinant domain 1 of the *P. vivax* sexual-stage antigen Pvs230 (Pvs230D1), expressed in *Pichia pastoris*, chemically conjugated to recombinant, non-toxic *Pseudomonas aeruginosa* ExoProtein A (rEPA) as a carrier protein to enhance its immunogenicity. Pvs230 is expressed on the surface of gametocytes in the human host and displayed on gametes in the mosquito host. Antibodies generated against Pvs230 disrupt parasite development within the mosquito midgut, thereby blocking transmission. Developed by the Laboratory of Malaria Immunology and Vaccinology (LMIV) at the National Institute of Allergy and Infectious Diseases (NIAID), Pvs230D1-EPA is formulated with the Matrix-M adjuvant and has been evaluated in Phase 1 clinical trials.
