Drug intelligence / Profile preview
Pvs25 DNA vaccine is a transmission-blocking vaccine (TBV) candidate designed to prevent the transmission of *Plasmodium vivax* malaria. It consists of a plasmid DNA vector encoding the Pvs25 antigen, a highly conserved 25 kDa ookinete surface protein of *P. vivax*. When administered, the vaccine induces host cells to express the Pvs25 antigen, eliciting a robust humoral immune response. When a mosquito feeds on an immunized individual, the ingested anti-Pvs25 antibodies target the Pvs25 protein on the surface of the parasite's ookinetes within the mosquito midgut, thereby preventing the development of oocysts and interrupting the malaria transmission cycle. The vaccine has been evaluated in preclinical studies, often delivered via in vivo electroporation to enhance immunogenicity.
