Overview

PVT101 is a pMHC nanomedicine developed by Parvus Therapeutics for the treatment of autoimmune conditions, specifically Type-1 diabetes and multiple sclerosis. It is based on the proprietary Navacim platform, which utilizes nanoparticles (typically iron oxide) functionalized with disease-specific peptide-major histocompatibility complexes (pMHC). These nanomedicines are designed to engage cognate T-cell receptors (TCRs) on autoreactive T-cells with high avidity. This interaction triggers the differentiation of these autoreactive cells into tissue-specific regulatory T-cells (Tregs), which subsequently suppress the autoimmune attack on target tissues—such as the insulin-producing beta cells in the pancreas or the myelin sheath in the central nervous system. This mechanism aims to restore antigen-specific immune tolerance without systemic immunosuppression. PVT101 is currently in the preclinical and IND-enabling phase of development.
