**PVX-2** is an investigational heterologous prime-boost therapeutic vaccine regimen developed by PapiVax for HPV16-positive low-grade cervical abnormalities, including ASC-US, ASC-H, LSIL, and CIN1. It comprises two separately administered intramuscular active vaccines: the HPV16 E7-targeted DNA plasmid vaccine pNGVL4a-Sig/E7(detox)/HSP70 and the recombinant HPV16 L2/E6/E7 fusion-protein vaccine TA-CIN. The regimen is intended to induce HPV16-specific cellular immunity, including E6/E7-specific CD4-positive and CD8-positive T-cell responses, promote clearance of HPV16-infected dysplastic cells, and generate L2-specific antibodies. In the Phase II safety run-in cohort, PVX-2 was well tolerated with no serious adverse events; 45% of evaluable participants were HPV16-negative at month 6 and 64% at month 12.
