Overview

PVX-410 + durvalumab is an investigational combination therapy consisting of a multi-peptide cancer vaccine (PVX-410) and a programmed death-ligand 1 (PD-L1) immune checkpoint inhibitor (durvalumab). PVX-410 is designed to stimulate the immune system by targeting multiple tumor-associated antigens, specifically XBP1, CD138, and CS1/SLAMF7, which are overexpressed in certain cancers such as triple negative breast cancer (TNBC) and multiple myeloma. The vaccine aims to induce cytotoxic T lymphocyte responses against these antigens. Durvalumab is a monoclonal antibody that blocks PD-L1, thereby enhancing T-cell mediated anti-tumor immunity by preventing tumor-induced immunosuppression. The combination has been studied in early-phase clinical trials as adjuvant therapy for HLA-A2+ patients with stage II/III TNBC following completion of standard treatment. The primary objectives have been safety and tolerability; secondary objectives include assessment of antigen-specific immune responses[1][2][3][4][5]. As of now, this combination remains investigational and has not received regulatory approval for any indication.
