**PVX1** is an investigational heterologous prime-boost therapeutic vaccine regimen for HPV16-associated malignancy. It consists of two intramuscular priming doses of the pNGVL4a-Sig/E7 (detox)/HSP70 plasmid DNA vaccine, which encodes a signal peptide, detoxified HPV16 E7 antigen, and HSP70, followed by an intramuscular boost with TA-HPV, a live recombinant vaccinia-virus vaccine expressing modified HPV16 and HPV18 E6 and E7 antigens. The regimen is being evaluated in healthy haploidentical donors to generate HPV-specific T cells for subsequent adoptive transfer or allogeneic transplantation strategies in recipients with recurrent or metastatic HPV16-associated cancer.
