PW 4153 is a potent, small-molecule dual inhibitor of the Pim kinase family (Pim-1, Pim-2, and Pim-3) and the Phosphoinositide 3-kinase (PI3K) pathway, specifically targeting the PI3K alpha isoform. Developed by Pathway Therapeutics, the compound was designed to overcome resistance mechanisms associated with single-pathway inhibition, as Pim kinases often provide alternative survival signals when the PI3K/Akt/mTOR pathway is blocked. Preclinical studies demonstrated that PW 4153 effectively inhibits protein synthesis, induces apoptosis, and suppresses the growth of various hematological and solid tumor cell lines. By simultaneously blocking these two critical signaling nodes, PW 4153 aims to achieve synergistic anti-tumor activity and prevent the compensatory upregulation of survival pathways that typically leads to treatment failure with monotherapies. However, clinical development of the compound appears to have ceased following the dissolution of its original developer.
