Overview

PXS-5131 is a small molecule, dual irreversible inhibitor of monoamine oxidase B (MAO-B) and semicarbazide-sensitive amine oxidase (SSAO), also known as vascular adhesion protein-1 (VAP-1). Developed by Pharmaxis, it is a fluoroallylamine derivative designed to treat neuroinflammatory and neurodegenerative disorders such as multiple sclerosis, Parkinson's disease, and Alzheimer's disease. PXS-5131 was engineered to overcome the limitations of its predecessor, mofegiline, by exhibiting improved physicochemical properties, a lack of phospholipidosis induction, and high selectivity over MAO-A and diamine oxidase (DAO). In preclinical models, PXS-5131 has demonstrated potent anti-inflammatory effects and the ability to reduce clinical symptoms in the experimental autoimmune encephalomyelitis (EAE) model of multiple sclerosis.
