PY159 is an afucosylated humanized IgG1 monoclonal antibody that specifically binds to the triggering receptor expressed on myeloid cells 1 (TREM1). Acting as an agonist, it crosslinks TREM1 and induces downstream signaling through the TREM1/DNAX Activating Protein of 12kDa (DAP12) complex. This activation reprograms immunosuppressive tumor-associated myeloid cells—such as monocytes, neutrophils, and macrophages—into proinflammatory states that promote antitumor immune responses. Preclinical studies demonstrated increased cytokine and chemokine production and upregulation of costimulatory markers following treatment with PY159. The drug was developed for use in solid tumors including breast cancer (HR+ HER2– and triple-negative), gastric cancer, pancreatic cancer, head and neck cancer, non-small cell lung cancer, gynecologic cancers, colorectal cancer, stomach adenocarcinoma, female genital neoplasms, adenocarcinoma of lung. Clinical development reached Phase 1 trials but has since been terminated[2][3][4][5][7][8].
