PY2 is a synthetic amino-pyrimidine analog of curcumin designed to address the inherent limitations of natural curcumin, such as its rapid degradation and poor bioavailability. By replacing the unstable 1,3-diketone moiety of curcumin with an amino-pyrimidine ring, PY2 achieves significantly enhanced chemical stability in physiological media. Preclinical evaluations have demonstrated that PY2 possesses potent anti-proliferative activity against several cancer cell lines, including prostate cancer (PC3 and LNCaP) and colorectal cancer (HCT116 and HT29). Its mechanism of action is believed to be pleiotropic, similar to curcumin, involving the modulation of multiple signaling pathways related to cell survival and apoptosis, although it shows improved cytotoxicity in specific models like PC3 cells.
