PY314 is a humanized IgG1 afucosylated monoclonal antibody that targets the triggering receptor expressed on myeloid cells 2 (TREM2), which is found on immunosuppressive, pro-tumorigenic tumor-associated macrophages (TAMs). By binding to TREM2, PY314 selectively depletes these TAMs in the tumor microenvironment, thereby promoting anti-tumor immunity. This mechanism shifts the balance from inhibitory M2-like TAMs toward more inflammatory M1-like TAMs and activates CD8 T-cells and natural killer cells to destroy tumors. Developed by Pionyr Immunotherapeutics, PY314 is being investigated for use in advanced solid tumors, including platinum-resistant ovarian cancer and other carcinomas. It has been evaluated as both a single agent and in combination with pembrolizumab in early-phase clinical trials[1][3][4][5][7].
