Drug intelligence / Profile preview
PYO2 is a lytic bacteriophage (specifically a myovirus belonging to the genus *Litunavirus*) that targets *Pseudomonas aeruginosa*. It specifically binds to the O-antigen of the lipopolysaccharide (LPS) on the outer membrane of *P. aeruginosa* to infect and lyse the bacterial cells. PYO2 is being investigated, particularly in combination with last-line antibiotics such as beta-lactams (e.g., aztreonam/avibactam) and polymyxin B, for the treatment of extensively drug-resistant (XDR) *P. aeruginosa* infections. Research has shown that resistance development to PYO2 selects for a bacterial subpopulation with increased outer membrane permeability, thereby restoring or enhancing sensitivity to beta-lactam antibiotics (collateral sensitivity).
