Pyricolib acetate (ZJU-06) is a potent, small-molecule inhibitor of the Menin-MLL (Mixed Lineage Leukemia) protein-protein interaction, developed by researchers at Zhejiang University. It is specifically designed for the treatment of acute leukemias, particularly those characterized by MLL rearrangements (MLL-r) or NPM1 mutations. By binding to the Menin protein, pyricolib disrupts its interaction with MLL fusion proteins or wild-type MLL, which are essential for the maintenance of a leukemogenic gene expression program. This disruption leads to the downregulation of critical oncogenic targets such as HOXA9 and MEIS1, subsequently inducing cell cycle arrest, differentiation, and apoptosis in leukemic cells. The drug is currently being evaluated in early-phase clinical trials to assess its safety and efficacy in patients with relapsed or refractory acute leukemia.
