Drug intelligence / Profile preview
The MGO-lowering cocktail is an experimental combination therapy consisting of pyridoxamine, hesperidin, and resveratrol. It is designed to target and reduce endogenous levels of methylglyoxal (MGO), a reactive glycolytic byproduct that accumulates during chronic hyperglycemia and aging, leading to advanced glycation endproducts (AGEs) and vascular complications. Mechanistically, pyridoxamine acts as a direct scavenger of MGO, while hesperidin and resveratrol synergistically enhance the enzymatic detoxification of MGO by upregulating glyoxalase 1 (GLO1) expression, likely via Nrf2 pathway activation. In preclinical models of type 1 diabetes, this cocktail has been shown to prevent the elevation of plasma MGO, improve aortic distensibility, and restore endothelial function, offering a promising therapeutic strategy for mitigating diabetes-related cardiovascular complications.
