Overview

Pyridoxine alpha-ketoglutarate (PAK) is a small molecule compound formed by the salt/complex of pyridoxine (vitamin B6) and alpha-ketoglutaric acid, a key intermediate in the Krebs cycle. It functions as a metabolic enhancer and a precursor for neurotransmitter synthesis. The pyridoxine component is converted into pyridoxal phosphate, which serves as a vital cofactor for enzymes such as glutamate decarboxylase (GAD) and aromatic L-amino acid decarboxylase (AADC), essential for the production of GABA, dopamine, and serotonin. The alpha-ketoglutarate component supports neuronal energy metabolism and nitrogen transport. PAK has been investigated for its potential in treating nervous system disorders, including tremors, convulsions, and Tourette syndrome. In clinical research, it is often evaluated in combination with magnesium pidolate, notably in a Phase IV trial in Spain sponsored by the Carlos III Health Institute to reduce tic severity in children. It is also commercially available as a dietary supplement.
