Overview

Pyrithiamine pyrophosphate is a synthetic derivative of pyrithiamine in which a pyrophosphate group is attached to the molecule. It acts as an antimicrobial agent and is structurally related to thiamine (vitamin B1). Pyrithiamine itself is known as an inhibitor of thiamine metabolism and can induce neurological symptoms similar to Wernicke-Korsakoff syndrome in animal models. The pyrophosphate form mimics thiamine pyrophosphate (TPP), the active coenzyme form of vitamin B1, but instead acts as an antagonist at TPP riboswitches found in bacteria and other organisms. By binding these riboswitches, it downregulates genes involved in thiamine metabolism and transport[2][3][7]. This mechanism underlies its antimicrobial activity.
