Pyronaridine + quinacrine + tilorone + vandetanib is a four-drug small molecule combination therapy being developed by Collaborations Pharmaceuticals for the treatment of COVID-19. This multi-agent approach is designed to inhibit SARS-CoV-2 by targeting both viral and host processes. The combination includes pyronaridine and quinacrine (antimalarials with lysosomotropic properties), tilorone (an antiviral inducer of interferon), and vandetanib (a multi-kinase inhibitor). The rationale for this combination involves modulating lysosomal function and inhibiting host kinases to disrupt viral entry and replication. The development program also identifies a target referred to as 'Pipro'. The drug is currently in the preclinical in-vivo stage of development.
