Pyronaridine + tilorone + quinacrine is a triple-drug combination therapy being developed by Collaborations Pharmaceuticals for the treatment of Marburgvirus Disease and Ebola Virus Disease. This combination utilizes three small molecule drugs—pyronaridine (an antimalarial), tilorone (a broad-spectrum antiviral), and quinacrine (an antimalarial/antiprotozoal)—which have demonstrated synergistic inhibitory effects against filoviruses. The mechanism of action primarily involves the inhibition of viral entry into host cells. This is achieved through multiple pathways, including the potential targeting of the viral glycoprotein (GP), inhibition of host cell proteases like cathepsin B required for viral fusion, and lysosomotropic effects that disrupt endosomal processing. The combination was identified through computational modeling and in vitro screening and is currently in preclinical development.
