Pyrotinib + paclitaxel is a combination therapy consisting of an irreversible pan-ErbB tyrosine kinase inhibitor and a microtubule-stabilizing taxane. Pyrotinib targets the intracellular kinase domains of EGFR, HER2, and HER4, inhibiting downstream signaling pathways like PI3K/Akt and MAPK. Paclitaxel interferes with microtubule dynamics during mitosis, leading to cell cycle arrest and apoptosis. This combination is primarily investigated for HER2-positive malignancies, including advanced or metastatic gastric and gastroesophageal junction cancers, where it aims to overcome resistance and provide synergistic anti-tumor activity.
