Overview

Pyrvinium is a small molecule anthelmintic drug primarily used for the treatment of pinworm (Enterobius vermicularis) infestations. It acts by interfering with glucose uptake in pinworms, leading to their death. Pyrvinium has been available for over 70 years and is considered safe and effective for its approved indication. In addition to its antiparasitic use, several forms of pyrvinium salts have been investigated for anticancer properties. Mechanistically, it inhibits mitochondrial function and Wnt/β-catenin signaling pathways, activates casein kinase 1α (CK1α), depletes cellular energy and autophagy processes, inhibits Akt signaling pathways, and shows activity against cancer stem cells. Despite promising preclinical data in various cancers—including colon, breast, lung, prostate cancer and hematological malignancies—pyrvinium has not yet been repurposed or approved as an anticancer agent[2][4][6][7][8].
