PYTX-004 is a modified, recombinant analogue of the opioid binding protein/cell adhesion molecule-like (OPCML), specifically engineered as an Fc fusion protein (r-OPCML-Fc). Developed by Papyrus Therapeutics, it is designed as a tumor suppressor biotherapeutic for cancer treatment. OPCML is a naturally occurring tumor suppressor that is frequently epigenetically silenced in many human cancers, leading to increased tumor invasion and metastasis. PYTX-004 acts as a protein replacement therapy to restore OPCML function at the cell surface, thereby reconstituting normal receptor tyrosine kinase (RTK) signaling and overcoming resistance to RTK inhibitors. Preclinical studies have shown efficacy in ovarian cancer models and potential for use in other solid tumors such as colorectal and gastric cancers. The drug works selectively on cancer cells deficient in OPCML without harming healthy cells[1][2][5].
