PYX-202 is a first-in-class, internalizing antibody-drug conjugate (ADC) targeting delta-like 1 homolog (DLK-1), a tumor-associated antigen expressed in a range of solid tumors but largely absent from normal adult tissues. PYX-202 uses a potent monoclonal antibody with high affinity for DLK-1 to drive efficient internalization, delivering a monomethyl auristatin E (MMAE) payload via a cleavable beta-glucuronide linker designed for increased stability and selective cleavage in tumor cells. The mechanism aims to maximize antitumor effect while minimizing off-target toxicity. Initially licensed by Pyxis Oncology from LegoChem Biosciences, its preclinical development was ultimately discontinued after toxicology studies[1][3][5][7].
