Drug intelligence / Profile preview
QC6352 is a small-molecule inhibitor originally developed by Celgene Quanticel Research as a potent, selective, and orally active pan-inhibitor of the lysine demethylase 4 (KDM4) family (specifically KDM4A, KDM4B, KDM4C, and KDM4D). It has been widely used as a chemical probe to study KDM4 biology and epigenetic regulation in various cancers, including breast, colorectal, and pediatric kidney cancers (such as Wilms tumor). However, a groundbreaking 2026 study published in *Nature Chemical Biology* revealed that QC6352's primary anticancer activity in glioblastoma stem cells is actually driven by the extremely potent inhibition of dihydroorotate dehydrogenase (DHODH), a mitochondrial enzyme essential for de novo pyrimidine biosynthesis, rather than KDM4 inhibition.
