Overview

QL1706 + oxaliplatin + capecitabine is a combination regimen consisting of a bispecific monoclonal antibody (QL1706) that targets both programmed cell death protein 1 (PD-1) and cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), plus the chemotherapy agents oxaliplatin and capecitabine. QL1706 is engineered as a mixture of a recombinant humanized IgG4 monoclonal antibody targeting PD-1 and a recombinant humanized IgG1 monoclonal antibody targeting CTLA-4, produced in a single cell line at a defined ratio to enhance correct assembly and minimize antibody mispairing. Oxaliplatin is a platinum-based chemotherapy that induces DNA crosslinks, inhibiting DNA replication and transcription, ultimately causing cell death. Capecitabine is an orally administered prodrug converted to 5-fluorouracil preferentially in tumor tissue, where it inhibits thymidylate synthase and disrupts DNA and RNA synthesis. This triple combination aims to provide the immune checkpoint blockade of QL1706 with the cytotoxic effects of standard chemotherapy, and is being clinically studied mainly in solid tumors such as advanced or metastatic cancers.
