QL335

QL335 is a bispecific T-cell engager (BiTE) developed by QLSF Biotherapeutics for the treatment of colorectal cancer (CRC). It is engineered in a 2+1 molecular format, featuring two high-affinity binding domains for Lymphocyte Antigen 6 Family Member G6D (LY6G6D) and one reduced-affinity binding domain for CD3 on T cells. LY6G6D is a GPI-anchored membrane protein that is highly expressed in microsatellite stable (MSS) colorectal cancer but exhibits minimal expression in normal tissues. By simultaneously bridging LY6G6D on tumor cells and CD3 on T cells, QL335 facilitates the recruitment and activation of T cells to selectively eliminate cancer cells. The 2+1 configuration is specifically designed to enhance tumor accumulation through avidity while mitigating non-specific T-cell activation in the periphery, potentially improving the safety profile.
