Overview

QLS-111 is a first-in-class, novel topical ophthalmic drug developed by Qlaris Bio for the treatment of primary open-angle glaucoma (POAG), ocular hypertension (OHT), and potentially normal tension glaucoma (NTG)[2][3][5]. It acts as an ATP-sensitive potassium channel opener, specifically targeting the relaxation of vascular and vascular-like tissues distal to the trabecular meshwork in the eye[1][2][5]. This mechanism reduces distal outflow resistance and lowers episcleral venous pressure (EVP), a key contributor to intraocular pressure (IOP) that is not directly addressed by current therapies[1][2][4]. By lowering EVP, QLS-111 provides significant IOP reduction both as monotherapy and in combination with standard treatments like latanoprost, showing additive effects in clinical trials[3][6]. The drug has demonstrated excellent safety and tolerability profiles with no serious adverse events or clinically meaningful hyperemia reported across multiple phase 2 studies[1][3][4].
