Overview

QLT-0267 is a small molecule inhibitor of integrin-linked kinase (ILK), an oncogenic protein that plays a critical role in cell survival, proliferation, and the regulation of centrosome clustering during mitosis. By inhibiting ILK, QLT-0267 induces centrosome declustering in cancer cells with supernumerary centrosomes, leading to aberrant mitotic spindle formation, mitotic arrest, and subsequent apoptosis. Research has demonstrated its potential in treating glioblastoma, particularly when used in combination with other agents like the Abl inhibitor imatinib, which can augment its effects on cell survival and mitotic progression. It has also been investigated for its ability to decrease cytosolic Abl levels and inhibit its localization to mitotic centrosomes.
