QP101 is a first-in-class dual-payload antibody-drug conjugate (ADC) targeting HER2, developed by Qurient Co., Ltd. in collaboration with Synaffix. It integrates a proprietary CDK7 inhibitor payload, which transcriptionally suppresses DNA damage response (DDR) genes, and a clinically validated topoisomerase I (TOP1) inhibitor payload (exatecan-based), enabling synergistic anti-tumor activity in HER2-expressing cancers. This design sensitizes tumors to TOP1i-induced DNA damage, allows lower TOP1i dosing to reduce toxicity, and expands potential use beyond traditional HER2-positive breast cancers to broader solid tumor indications, including earlier treatment lines.[1][2]
