QR-1123 is a first-in-class investigational antisense oligonucleotide designed to treat autosomal dominant retinitis pigmentosa (adRP) caused by the P23H mutation in the rhodopsin (RHO) gene. The drug works by specifically binding to the mutant RHO mRNA, leading to allele-specific knockdown of the mutant transcript via RNase H-mediated cleavage, thereby inhibiting production of the toxic mutated rhodopsin protein without affecting normal RHO mRNA. It is administered through intravitreal injection and was originally developed by Ionis Pharmaceuticals using their proprietary antisense technology before being in‑licensed by ProQR Therapeutics for further development[1][2][3][4][6].
