QTX3046 + cetuximab is an investigational combination therapy currently in Phase 1 clinical development for solid tumors harboring KRAS G12D mutations. QTX3046 is a chemically distinct, orally bioavailable, non-covalent, allosteric small molecule that functions as a KRAS G12D-selective dual ON/OFF state inhibitor. It targets oncogenic KRAS G12D by direct inhibition, disrupting its active/inactive state cycling. Cetuximab is an EGFR-targeting monoclonal antibody used to block the epidermal growth factor receptor (EGFR) pathway. The rationale for the combination is that allosteric KRAS inhibition paired with EGFR blockade may synergistically enhance tumor response in cancers such as colorectal and pancreatic adenocarcinomas. Both agents are administered concomitantly in ongoing clinical studies, where endpoints include safety, pharmacokinetics, and antitumor activity[1][4][5].
